The role of the oxidative pentose phosphate (PP) pathway in the dormancy-breaking of cocklebur (Xanthium pennsylvanicum Wallr.) seeds was investigated. D-[1-14C]-glucose or D-[6-14C]-glucose was fed to dormant and non-dormant lower seeds or to their axial or cotyledonary segments which were imbibed for different durations, and C6/C1 ratios of respired 14CO2 as an index of the PP pathway activity were calculated. Contrary to expectation, there was no significant difference in the C6/C1 ratios between the dormant and non-dormant seeds or segments during a water imbition period of 24 h, although the PP pathway actually operated already in an early stage of water imbibition. Also concerning the activities of G6PDH and 6PGDH, the key enzymes of this pathway, no difference between the dormant and non-dormant seeds was found. It was thus concluded that, unlike other seeds, there is no contribution of the PP pathway to the regulation of dormancy of the cocklebur seed.  相似文献

AIMS: To evaluate the effect of water activity (a(w) 0.98-0.89, adjusted with glycerol, sorbitol, glucose, or NaCl) and temperature (5-25 degrees C) on the lag phase and radial growth rate (mm day(-1)) of the important citrus spoilage fungi, such as Penicillium italicum and Penicillium digitatum grown in potato dextrose agar (PDA) medium. To select, among models based on the use of different solutes, a model fitting accurately the growth of these species in relation to a(w) and temperature. METHODS AND RESULTS: Extensive data analyses showed for both Penicillium species a highly significant effect of a(w), temperature, solutes and their interactions on radial growth rate (P < 0.0001). Radial growth rate was inhibited and the lag phase (i.e. the time required for growth) lengthened as the a(w) of the medium decreased. NaCl appeared to causes the greatest stress on growth when compared with other nonionic solutes. Penicillium italicum stopped growing at 0.96 a(w) and P. digitatum at 0.93 a(w). Under the dry conditions where growth was observed, P. italicum grew faster than P. digitatum at low temperature and P. digitatum remained more active at ambient temperature. Multiple regression analysis applied to the square roots of the growth rates observed in the presence of each solute showed that both the 'glycerol model' and the 'sorbitol model' yielded a good prediction of P. italicum growth and the 'sorbitol model' gave an accurate fit for P. digitatum growth, offering high-quality prediction within the experimental limits described. CONCLUSIONS: Mathematical models describing and predicting, as a function of a(w) and temperature, the square root of the radial growth rate of the agents responsible for blue and green decays are important tools for understanding the behaviour of these fungi under natural conditions and for predicting citrus fruit spoilage. SIGNIFICANCE AND IMPACT OF THE STUDY: Implementation of these results should contribute towards a more rational control strategy against citrus spoilage fungi.  相似文献

Evolution of HCN from both rice ( Oryza sativa ) and cocklebur ( Xanthium pennsylvanicum ) seeds increased during a pre-germination period and preceded the evolution of (C2H4). These two species were adopted as the representatives of starchy and fatty seeds, respectively. Ethylene promotes seed germination of many species. However, HCN evolution declined abruptly when the radicles emerged and before the peak in C2H4 evolution. More-over, both rice and soybean ( Glycine max ) seeds showed some activity of β-cyanoalanine synthase (CAS, EC 4.4.1.9) even in the unimbibed dry state. The activities of CAS in the lower seed of cocklebur and in soybean seeds increased rapidly after emergence of the radicle. However, the CAS of rice seeds, with high activity in the dry state, exhibited a bimodal change, gradually decreasing until radicle emergence had occurred, but then increaing. It is thus likly that HCN evolution during initial imbibition may be derived from cyanogenic reserves and controlled by both pre-existing and subsequently-developing CAS. The exogenous application of C2H4 stimulated the activities of CAS in both rice and upper cocklebur seeds and reduced their cyanogen contents. Therefore, the decline of HCN evolution after germination seems to be due to the increased activities of CAS by endogenously produced C2H4.  相似文献

Breaking dormancy in some seeds requires a period of dry storage. In the seeds of Xanthium pennsylvanicum Wallr., the process of afterripening proceeds optimally at water contents between 7 and 14%: this range of dehydration can be identified with water binding region 2, in which water is bound with low enthalpy. At water contents below 7%. Seeds remained primarily dormant over 3 years. Attempts to alter the afterripening with atmospheres of elevated nitrogen showed no effect. and with oxygen there was no consistent effect. There were no changes is osmotic value of the seed sap, or in its sugar or amino acid contents. We speculate that afterripening in Xanthium may involve some nonenxymatic reactions which remove substances which inhibit germination. Candidates for these reactions include the Amadori and Maillard reactions.  相似文献

Energy-use efficiency and energy assimilation, investment and allocation patterns are likely to influence plant growth responses to increasing atmospheric CO2 concentration ([CO2]). Here, we describe the influence of elevated [CO2] on energetic properties as a mechanism of growth responses in Xanthium strumarium. Individuals of X. strumarium were grown at ambient or elevated [CO2] and harvested. Total biomass and energetic construction costs (CC) of leaves, stems, roots and fruits and percentage of total biomass and energy allocated to these components were determined. Photosynthetic energy-use efficiency (PEUE) was calculated as the ratio of total energy gained via photosynthetic activity (Atotal) to leaf CC. Elevated [CO2] increased leaf Atotal, but decreased CC per unit mass of leaves and roots. Consequently, X. strumarium individuals produced more leaf and root biomass at elevated [CO2] without increasing total energy investment in these structures (CCtotal). Whole-plant biomass was associated positively with PEUE. Whole-plant construction required 16.1% less energy than modeled whole-plant energy investment had CC not responded to increased [CO2]. As a physiological mechanism affecting growth, altered energetic properties could positively influence productivity of X. strumarium, and potentially other species, at elevated [CO2].  相似文献

The hexaploid herbaceous species Primula glaucescens Moretti and Primula spectabilis Tratt. (2n = 6x = 66) are two endemics of the Italian Pre-Alps protected by national and international laws. In order to plan conservation strategies for natural populations and to study the influence of the latest glaciation on them we developed a set of microsatellites markers for the endangered Primula species: ten primer pairs allowed analysis of polymorphic disomic loci in P. glaucescens samples and seven of them also amplified polymorphic disomic markers in P. spectabilis. 为了明确不同储藏方式和储藏时间对三峡水库消落区4种1年生植物稗(Echinochloa crusgalli)、苍耳(Xanthium sibiricum)、合萌(Aeschynomene indica)和水蓼(Polygonum hydropiper)种子萌发能力的影响,筛选出有利于其萌发的最佳储藏方式及时间,采取5种储藏方式(室温、冷藏、冷冻、干沙、湿沙)、8个储藏时间段(30、60、90、120、150、180、210、240d)对种子进行储藏实验。在光照培养箱昼25℃/夜20℃和周期性光照(昼12h/夜12h,光强100μmol·m-·2s-1)条件下进行种子萌发,每次萌发持续时间为30d。结果显示:(1)稗、苍耳和水蓼的种子在湿沙储藏条件下萌发起始时间缩短,种子萌发率、萌发指数高于其它储藏方式;湿沙储藏条件下,稗的种子储藏90-180d萌发率达到80%以上,苍耳的种子储藏180d萌发率达(64±2)%,水蓼的种子储藏30-240d萌发率均超过90%。(2)冷冻储藏能有效地解除合萌种子休眠,显著提高其种子萌发率和发芽指数;冷冻储藏90-180d合萌的种子萌发率均超过80%,之间没有显著差异。(3)果皮是限制苍耳种子萌发的主要因素之一,而去除果皮对合萌种子萌发没有显著影响。研究结果建议:采用种子来进行消落区植被恢复和重建时,稗、苍耳和水蓼的最佳储藏方式是湿沙储藏,合萌的最佳储藏方式是冷冻储藏。结合三峡水库水位调度原则,在适宜的储藏方式下,本实验中储藏120-180d的种子能够应用于消落区实地播种中。  相似文献

Plant responses to elevated CO2 concentrations ([CO2]) may be regulated by both accelerated ontogeny and allocational changes as plants grow. However, isolating ontogeny‐related effects from age‐related effects are difficult because these factors are often confounded. In this study, the roles of age and ontogeny in photosynthetic responses to elevated [CO2] were examined on Xanthium strumarium L. grown at ambient (365 µmol mol?1) and elevated (730 µmol mol?1) [CO2]. To examine age‐related effects, six cohorts were planted at 5‐day intervals. To examine ontogeny‐related effects, all plants were induced to flower at the same time; ontogeny in Xanthium is relatively unaffected by growth in elevated [CO2]. Growth in elevated [CO2] increased net photosynthetic rates by approximately 30% throughout vegetative growth (i.e. active carbohydrate sinks), approximately 10% during flowering (i.e. minimal sink activity), and approximately 20% during fruit production (i.e. active sinks). At the harvest, the ratio of source to sink tissue significantly decreased with increasing plant age and was correlated with leaf soluble sugar concentration. Leaf soluble sugar concentration was negatively correlated with the relative photosynthetic response to elevated [CO2]. These results suggest that age and ontogeny independently affect photosynthetic responses to elevated [CO2] and the effects are mediated by reversible changes in source : sink balance.  相似文献
